(AEON) announced positive results from a pilot forced degradation study comparing ABP-450 with BOTOX (onabotulinumtoxinA), adding to the analytical evidence supporting the products' comparability. Forced degradation testing assesses how biologic molecules respond to stress conditions such as heat and oxidation, providing additional information on structural similarities beyond testing of unstressed material. Under thermal stress, the two products showed comparable rates of potency loss, with both reaching approximately half of their initial observed potency by day 14, based on an LD50 assay. Under oxidative stress, ABP-450 and BOTOX were exposed to two concentrations of hydrogen peroxide. Both products showed similar dose-dependent oxidation patterns at the evaluated methionine sites, as measured by mass spectrometry peptide mapping. ABP-450 (prabotulinumtoxinA) is a 900-kDa botulinum toxin type A complex that AEON is developing as a biosimilar to BOTOX for therapeutic indications. The pilot study was not powered for formal statistical comparison, and the company said formal conclusions will depend on a planned multi-lot comparability study, which it expects to initiate in the first half of 2027. The latest findings build on previously reported analytical data, including 100% amino-acid sequence identity across analyzed regions and comparable potency and functional characteristics between ABP-450 and BOTOX. AEON is pursuing the FDA's 351(k) biosimilar pathway and plans to incorporate the findings into its broader analytical package supporting a potential 351(k) BLA filing for ABP-450. AEON closed Thursday's trade at $0.23, down 1.70%. In the pre-market trading, shares are up 41.11% at $0.34.
